Abstract

In a recent report concerning the reversibility by chlorpromazine of psychoses induced by means of mescaline and d-lysergic acid diethylamide (LSD-25), careful attention was given to the clinical and electroencephalographic changes.1 Although the clinical changes of this reversal effect were immediate and striking, the conventional electroencephalographic changes were minimal, and it was difficult to assess them. It was noted in another recent study that mescaline and LSD-25 failed to activate temporal-lobe epilepsy either clinically or electroencephalographically.2

Because of these factors, it was hoped that depth electrographic studies might be fruitful in elucidating the effects of these drugs. Adrenochrome is a metabolic product of adrenaline and is involved in the formation of the skin pigment . It was further investigated in 1952 as part of clinical studies to understand schizophrenia and how to combat it. It is closely related to adrenolutin, which has similar properties. It belongs to the chemical group of aminochromes.

In the early 1950s, all sorts of means to combat mental illnesses were researched. A. Hoffer and H. Osmond presented u. a. hypothesized that the body's own adrenochrome, which was discovered in the 1940s, fueled the “imagination” of schizophrenic personalities and thus made a decisive contribution to the clinical picture. [4] They referred to this hypothesis as the adrenochrome hypothesis, which should expand the current and recognized dopamine hypothesis and lead to new treatment methods.

The hallucinogenic effects of adrenochrome were also discovered in the course of their studies on the intervention in the body's adrenochrome household to treat schizophrenic personalities. This effect seemed very similar to that of LSD and mescaline, if not as potent. Hoffer and Osmond then hypothesized that LSD and mescaline boost the body's production of adrenochrome and are therefore only responsible for its hallucinogenic effects. They described their studies on hallucinogenic effects in 1967 in their joint textbook "The Hallucinogens".

Bernard W. Aronson served four U.S. Presidents in senior positions. In his remarks at the February 4, 2016 White House celebration of the 15th anniversary of Plan Colombia, President Barack Obama said, "I especially want to recognize someone who's played a pivotal role in the peace process. Aronson left government service in 1993, joining Goldman Sachs as an International Advisor for Latin America. Humphry Fortescue Osmond (1 July 1917 – 6 February 2004) was an English psychiatrist who expatriated to Canada, then moved to work in the United States. He is known for inventing the word psychedelic and for his research into interesting and useful applications for psychedelic drugs. Osmond also explored aspects of the psychology of social environments, in particular how they influenced welfare or recovery in mental institutions.

After the war, Osmond joined the psychiatric unit at St George's Hospital, London where he rose to become senior registrar. His time at the hospital was to prove pivotal in three respects, firstly it was where he met his wife Amy "Jane" Roffey who was working there as a nurse, secondly he met Dr John Smythies who was to become one of his major collaborators, and thirdly he first encountered the drugs that would become associated with his name (and his with theirs): LSD and mescaline.

At Weyburn, Osmond recruited a group of research psychologists to turn the hospital into a design-research laboratory. There, he conducted a wide variety of patient studies and observations using hallucinogenic drugs, collaborating with Abram Hoffer and others. In 1952, Osmond related the similarity of mescaline to adrenaline molecules, in a theory which implied that schizophrenia might be a form of self-intoxication caused by one's own body. He collected the biographies of recovered schizophrenics, and he held that psychiatrists can only understand the schizophrenic by understanding the rational way the mind makes sense of distorted perceptions. He pursued this idea with passion, exploring all avenues to gain insight into the shattered perceptions of schizophrenia, holding that the illness arises primarily from distortions of perception. Yet during the same period, Osmond became aware of the potential of psychedelics to foster mind-expanding and mystical experiences.

In 1953, English-born Aldous Huxley was long-since a renowned poet and playwright who, in his twenties, had gone on to achieve success and acclaim as a novelist and widely published essayist. He had lived in the U.S. for well over a decade and gained some experience screenwriting for Hollywood films. Huxley had initiated a correspondence with Osmond. In one letter, Huxley lamented that contemporary education seemed typically to have the unintended consequence of constricting the minds of the educated—close the minds of students, that is, to inspiration and to many things other than material success and consumerism. In their exchange of letters, Huxley asked Osmond if he would be kind enough to supply a dose of mescaline.

Osmond's interests included the application of Jung's Typology of personality to group dynamics. He and Richard Smoke developed refinements of Jung's typology and applied them to analysis of the presidents and other world figures. Osmond also studied parapsychology.

Later, Osmond became director of the Bureau of Research in Neurology and Psychiatry at the New Jersey Neuro Psychiatric Institute (NJNPI) in Princeton, where he collaborated with Bernard Aaronson in hypnosis experiments. Still later, he became a professor of psychology at the University of Alabama in Birmingham. Osmond co-wrote eleven books and was widely published throughout his career.

Julius Axelrod (May 30, 1912 – December 29, 2004)[1] was an American biochemist. He won a share of the Nobel Prize in Physiology or Medicine in 1970 along with Bernard Katz and Ulf von Euler.[2][3][4][5] The Nobel Committee honored him for his work on the release and reuptake of catecholamine neurotransmitters, a class of chemicals in the brain that include epinephrine, norepinephrine, and, as was later discovered, dopamine. Axelrod also made major contributions to the understanding of the pineal gland and how it is regulated during the sleep-wake cycle.

In 1946, Axelrod took a position working under Bernard Brodie at Goldwater Memorial Hospital. The research experience and mentorship Axelrod received from Brodie would launch him on his research career. Brodie and Axelrod's research focused on how analgesics (pain-killers) work. During the 1940s, users of non-aspirin analgesics were developing a blood condition known as methemoglobinemia. Axelrod and Brodie discovered that acetanilide, the main ingredient of these pain-killers, was to blame. They found that one of the metabolites also was an analgesic. They recommended that this metabolite, acetaminophen (paracetamol, Tylenol), be used instead.

Catecholamine research

In 1949, Axelrod began work at the National Heart Institute, forerunner of the National Heart, Lung, and Blood Institute (NHLBI), part of the National Institutes of Health (NIH). He examined the mechanisms and effects of caffeine, which led him to an interest in the sympathetic nervous system and its main neurotransmitters, epinephrine and norepinephrine. During this time, Axelrod also conducted research on codeine, morphine, methamphetamine, and ephedrine and performed some of the first experiments on LSD. Realizing that he could not advance his career without a PhD, he took a leave of absence from the NIH in 1954 to attend George Washington University Medical School. Allowed to submit some of his previous research toward his degree, he graduated one year later, in 1955. Axelrod then returned to the NIH and began some of the key research of his career.

Axelrod received his Nobel Prize for his work on the release, reuptake, and storage of the neurotransmitters epinephrine and norepinephrine, also known as adrenaline and noradrenaline. Working on monoamine oxidase (MAO) inhibitors in 1957, Axelrod showed that catecholamine neurotransmitters do not merely stop working after they are released into the synapse. Instead, neurotransmitters are recaptured ("reuptake") by the pre-synaptic nerve ending, and recycled for later transmissions. He theorized that epinephrine is held in tissues in an inactive form and is liberated by the nervous system when needed. This research laid the groundwork for later selective serotonin reuptake inhibitors (SSRIs), such as Prozac, which block the reuptake of another neurotransmitter, serotonin. .. In 1958, Axelrod also discovered and characterized the enzyme catechol-O-methyl transferase, which is involved in the breakdown of catecholamines.

Pineal gland research

Some of Axelrod's later research focused on the pineal gland. He and his colleagues showed that the hormone melatonin is generated from tryptophan, as is the neurotransmitter serotonin. The rates of synthesis and release follows the body's circadian rhythm driven by the suprachiasmatic nucleus within the hypothalamus. Axelrod and colleagues went on to show that melatonin had wide-ranging effects throughout the central nervous system, allowing the pineal gland to function as a biological clock

For example, during the First World War, when he was called to Royal Engineers Experimental Station (near Salisbury) to carry out experiments on asphyxiating gas, he exposed himself to an atmosphere of poisonous hydrogen cyanide. On another occasion he remained for seven days in a glass chamber in order to calculate the minimum quantity of oxygen required for the survival of the human organism, and another time he exposed himself to such a low temperature that he collapsed into unconsciousness.

Between 1902 and 1905 he was a Governor of Leighton Park School, the Quaker School in Reading. From 1925 to 1937 he held the chair of physiology at Cambridge. His final research, begun in 1933, concerned fetal respiration.

He was knighted in 1935.[4] He was elected a Foreign Honorary Member of the American Academy of Arts and Sciences in 1938.[6]

During the first years of the Second World War he was again summoned to Porton Down to consult on chemical weapons.

The dehydrogenase catalysed the transfer of hydrogen from the substrate to the coenzyme; in turn the reduced coenzyme reduced the carrier, and finally reduced carrier reacted with molecular oxygen. The function of the ketone fixative consisted in binding the keto-acid formed by the oxidation of either lactic or malic acid. The product of oxidation

in both cases completely arrested the catalytic oxidation unless removed by the ketone reagent.

Adrenaline, flavin and flavoprotein were the only substances occurring in animal tissues which were found capable of acting as carriers in the lactic and malic systems. The mechanism of the flavin and flavoprotein effects was perfectly clear. These substances were alternately reduced by the coenzyme and oxidized by molecular oxygen. This simple interpretation however failed to account for the adrenaline effect. The experiments of Green & Brosteaux [1936] and Green [1936, 2] showed clearly that the action of adrenaline as an oxidation carrier involved a complicated mechanism.

Derek Richter (14 January 1907 – 15 December 1995), English neuroscientist, was one of the founding fathers of the science of brain chemistry. Identifying monoamine oxidase and demonstrating its function in the brain early in his career, he became a prime mover in his field. In 1968 he was involved in the foundation of the British Brain Research Organization (BRA). Prior to this he helped to establish the Mental Health Research Fund, later renamed The Mental Health Foundation,(1949).[1] He also established the Journal of Neurochemistry (1956), of which he was one of the chief editors from 1956 to 1969. He was involved in the foundation of the International Brain Research Organization (IBRO) (1960), of which he became secretary-general, and the International Society for Neurochemistry (ISN) (1967). After leading a research unit at Whitchurch Psychiatric Hospital (1947–1960), he became director of the Medical Research Council Neuropsychiatric Research Unit in Carshalton (1960–1971).[2] Throughout his life he promoted international collaboration in research.

In 1931 he gained a PhD (magna cum laude) for the autoxidation of aldehydes. He then moved to Sir Frederick Gowland Hopkins Biochemical Laboratory at Cambridge alongside Nobel laureates Ernst Chain, Richard Synge, Frederick Sanger, Hans Krebs and Archer Martin. This was a defining period, working with Hermann Blaschko on monoamines that have great physiological interest. A year before the outbreak of World War II, he took a post at the Maudsley Hospital to work on amphetamines. Married to Beryl Ailsa Griffiths, with whom he had three children, he set up a wartime research laboratory for treating shell-shock in the Mill Hill Emergency Hospital.

..In 1947 Richter received a Rockefeller Foundation grant to set up a laboratory in the Whitchurch Psychiatric Hospital, Cardiff. Four years later, with one of the first Geiger counters to measure radioisotopes in the country, he and RMC Dawson had isolated nuclei from the cells of the cerebral cortex and demonstrated changes in brain metabolism with changes in brain function. With Linford Rees he carried out early work on biochemical changes concerning epilepsy and electroconvulsive therapy (ECT) and collaborated with W. Grey-Walter in EEG studies. With Miki Gaitonde, he worked on the metabolic activity of proteins of the brain.

At Carshalton, Richter gathered a multidisciplinary international group of scientists with a global approach to their research. He became involved with the World Health Organization, and Amnesty International. In retirement he was elected a fellow of the Royal College of Psychiatrists, and wrote a series of humanitarian books.

Concern for the childless women prompted him, through medical colleague Bertold Wiesner to become one of the first sperm donors at Mary Barton (Wiesner's wife)'s insemination clinic in London. Some of these children have been identified by DNA markers.[5][6] Between April 1945 and 1951 he made weekly contributions at a time when AID was strictly anonymous and very controversial.[7][8] It was a precursor to modern developments in reproductive technology such as IVF, that are leading to changes in the structure of families.

He continued to write and edit enlightened books, gardened passionately and joined his second wife, Molly Bullock, in establishing South Lodge, a refuge in Epsom for discharged mental patients.
