Like all the cases referenced in pizzagate - EVERY one - this is not strictly speaking a coloboma. It's a dark spot on the surface of an iris with normal shape. This is called iris nevus or iris freckle. It's a common finding, occurring in up to 5% of the population, although not always large or prominent.

Many people will hate this blasphemy that I write; probably it will get downvotes. These people would be surprised to read the interview with Madeleine McCann's mother in which she says that Madeleine actually had a freckle on her eye herself. After first calling it coloboma she corrects herself and says freckle. See towards the end of this transcript from CNN McCann interview:

But mccann's credibility is it not high in other matters so no doubt people will continue believing all the victims have coloboma and make up reasons why, even though geneticists do not fully know why or understand all implications.

Focusing too much on weird looking eyes may be fun but it is likely a time waster. Just pointing this out (again) to be helpful.
